Flu antibody?s 'one-handed grab' may boost effort toward universal vaccine, new therapies

ScienceDaily (Sep. 12, 2012) ? Scientists from The Scripps Research Institute and Sea Lane Biotechnologies have solved the co-crystal structure of a human antibody that can neutralize influenza viruses in a unique way. The antibody recognizes the crucial structure that flu viruses use to attach to host cells, even though previously this structure had been thought too small for an antibody to grab effectively. The immune protein manages to hit this precise spot by using just a small part of its target-grabbing apparatus. In so doing, it can neutralize a broad range of dangerous flu viruses.

"This highly focused binding to the receptor binding site using only a single loop on the antibody has never been seen before, and it's really fascinating; it gives us some good ideas about designs for vaccines and therapies," said Ian A. Wilson, the Hansen Professor of Structural Biology at Scripps Research. Wilson was senior investigator of the study along with Ramesh R. Bhatt of Sea Lane Biotechnologies. The report appears online ahead of print on September 16, 2012, in the journal Nature.

The Power of Large Numbers

Sea Lane Biotechnologies, advised by Richard Lerner, the Lita Annenberg Hazen Professor of Immunochemistry at Scripps Research, began by collecting bone marrow from patients who had been exposed to certain key strains of flu. Because the bone marrow is a "fossil record" of all the antibodies a person has ever made, those at Sea Lane felt confident that the antibodies they were looking for would be there. Sea Lane's efforts even went as far as to include advertised appeals on Craigslist for individuals who had certain strains of the flu. Using the internationally and locally collected bone marrow, Sea Lane generated a "comprehensive flu library" of billions of antibodies.

Sea Lane Biotechnologies scientists, led by Bhatt, isolated the unusual new antibody, which they dubbed C05, by screening this enormous library for antibodies that could bind to proteins from a variety of influenza A viruses -- the most dangerous family of flu viruses.

C05 also protected cells in the lab dish from infection by these viruses. In mice, relatively low doses of C05 prevented infections despite influenza A exposures that would have been lethal. The antibody worked as a therapy, too, rescuing 100 percent of mice when administered up to three days after a flu infection had begun.

An Unusual Grip for a Tricky Target

Further tests revealed a curious property of C05. Almost uniquely among broadly neutralizing antibodies against influenza A, it specifically recognizes and blocks the part of the flu virus that mediates viral attachment to host cells. Known as the "receptor binding site" (RBS), this viral site is located on the heads of viral hemagglutinins, spiky structures of sugar and protein that coat the viral envelope.

The RBS is a key functional site on flu viruses and is relatively exposed to the immune system compared to other viral regions. It would be an ideal target for antibodies, except that it is quite small compared to an antibody's usual grip area.

An antibody binds targets using two arm-like structures, each of which has six protein fingers or loops. To get a good grip on the flu RBS with some or all these loops, a typical antibody has to grab not only the RBS, but also some of the surrounding structures in the head, which vary from one flu strain to another. Thus, an antibody that does get a firm grip on this region for one flu strain generally will lose that grip once the strain mutates. "This is why universal flu vaccine strategies in recent years have been focused more on the hemagglutinin stem than on the head," noted Damian C. Ekiert, formerly of Wilson's laboratory at Scripps Research and now of University of California, San Francisco. Ekiert was first author of the paper with Arun K. Kashyap of Sea Lane Biotechnologies.

Wilson's laboratory specializes in the use of X-ray crystallography and other techniques to determine precisely where and how such antibodies bind to their viral targets. Using these methods, his team found that C05 effectively avoids grabbing the hypervariable regions around the flu RBS. Instead it uses a single elongated protein loop to reach in and make a "one-handed" -- or "one-fingered" -- grab of the RBS itself. The antibody apparently works best when two of these active loops, one on each arm, grab two viral RBSs on separate hemagglutinins. "It looks like these antibodies need to cross-link two hemagglutinins to have maximum effect," said Wilson.

Potentially Useful

The RBS has such an important function that it does not change much from strain to strain -- and thus C05 can neutralize a broad range of dangerous influenza A viruses, including H1, H2, H3, and H9 subtypes.

The fact that C05 can make such a precision grab suggests that it and perhaps even more potent antibodies could contribute to antibody-based therapies for severe influenza infections. A universal flu vaccine also might be significantly more effective if it could be designed to elicit such antibodies in people.

"If we can figure out how to induce this sort of antibody in a vaccine, we would have something that's very useful," said Lawrence Horowitz, CEO of Sea Lane Biotechnologies.

In addition to Wilson, Bhatt, Ekiert, Kashyap, and Horowitz, authors of the paper, "Neutralization of Influenza A viruses by insertion of a single antibody loop into the receptor binding site," included Michael A. Ward of Scripps Research.

Town moves toward energy excise tax participation

Braselton is following one of its county?s plans to potentially impose a new energy excise tax on manufacturers ? a move that could offset some anticipated revenue drop in local sales tax.

By the end of this year, elected officials in cities and counties across Georgia must decide if they will collect a new, optional excise tax on energy from manufacturers. The move comes after Gov. Nathan Deal signed a new state law in April that exempts manufacturers from paying state and local sales tax for energy.

While the measure was intended to encourage economic development, governments across Georgia are now discussing how they?ll address the dip in sales tax revenue from the tax break. The new state law gives city and counties the option to impose an excise tax to recoup some of that lost revenue.

?Those are revenues that the county has been obviously anticipating collecting and has done their projects based on full collection,? town manager Jennifer Dees told the Braselton Town Council on Thursday.

But the big question mark in the revamped sales tax law is how much it will affect local governments.

?No one knows how much it is, however,? Dees said. ?The (Georgia) Department of Revenue cannot tell us how much it is. The energy companies can?t tell us how much it is. All that anyone really knows is that it?s dwindling revenue and they don?t know how much it is and they?ve got to decide if they can lose any.?

With the new state law, the energy exemption on sales tax will be phased in over a four-year period until fully implemented in 2016. Likewise, the excise tax imposed by local governments will follow the phase-in process.

Even if with the excise tax, manufacturers would still be paying less in energy cost because the state?s sales tax rate is four percent. Depending on how they currently impose sales taxes, counties and cities can have an energy tax rate of up to two percent.

Those communities with just one form of sales tax ??such as SPLOST (Special Purpose Local Option Sales Tax) or LOST (Local Option Sales Tax) ??can get one percent in energy excise tax. Those communities with two sales tax programs ??such as SPLOST and LOST ? may get up to two percent. The excise tax doesn?t apply to Education SPLOST, according to Dees.

For now, counties across Georgia are considering if they will impose the new tax on manufacturers.

Counties have until Dec. 31 to impose the energy excise tax, although they have the option of implementing anytime after that. Those counties that adopt the new tax before Dec. 31 must have an intergovernmental agreement with the cities in their county to collect revenue. Those cities located in counties that choose not to collect the excise tax by Dec. 31 have the option to implement the new tax on manufacturers within their city limits.

As a town located in four counties ??Jackson, Barrow, Gwinnett and Hall ??Braselton may potentially have to adopt several intergovernmental agreements.

Excise tax revenue will be based on existing distributions among counties and cities for SPLOST and LOST. Money from the excise tax may be used in a government?s general fund budget ??not just capital projects, as SPLOST requires.

On Monday, the Braselton Town Council approved an intergovernmental agreement with Hall County for the new energy excise tax.

Gwinnett County is moving forward with the excise tax and will meet with city officials to discuss the proposal, according to Dees. Barrow County is also getting intergovernmental agreements with its cities.

However, Jackson County officials have opted to not collect the excise tax at this time until they know how much revenue the county will lose, Dees said.

Braselton would then have the option to collect the excise tax from manufacturers that are located in the Jackson County portion of the town, she explained. In that case, the town has about three such manufacturers.

Other Business
In other business, the Braselton Town Council:
?met in a closed-door meeting for an hour and 15 minutes on Monday to discuss land acquisition. When the meeting was opened to the public, the council voted to authorize the town manager and town attorney to acquire the property discussed in executive session by ?any means necessary.?

?approved its Capital Improvement Element (CIE) and Short Term Work Program (STWP) updates to the town?s comprehensive plan. The CIE is required because the town collects impact fees on new developments. The projects in the CIE and the STWP are mostly all projects that are in the works or have been discussed by the council. The only changes to the documents was the removal of projects that have been completed. The town council agreed to transmit the CIE and STWP to the Northeast Georgia Regional Commission.

Chinese media says vice president appears in public

BEIJING (Reuters) - China's leader-in-waiting Xi Jinping appeared in public on Saturday for the first time in about two weeks, visiting a Beijing university in what appeared to be an effort to dispel rumors of serious illness and a troubled succession.

In a brief English-language report, the Xinhua news agency said Vice President Xi "arrived at China Agricultural University Saturday morning for activities marking this year's National Science Popularisation Day".

A single picture on the government's website (www.gov.cn) showed Xi, with a slight smile and wearing a black informal jacket over a white shirt, walking around the university.

Reuters had reported that Xi was likely to make an appearance on Saturday.

Sources have told Reuters that Xi hurt his back while swimming earlier this month and that he had been obeying doctors' orders to get bed rest and undergo physiotherapy.

A Reuters reporter at the university saw a man with sleek black hair wearing a white shirt -- who from a distance looked like Xi -- getting loud applause as he stepped out of the building housing an exhibition and raised his arms up and down twice in a gesture of vigour.

There was a light security presence around the university, but a building housing a science exhibition was closed off by police and plain clothes guards.

Hundreds of students applauded, some shouting "Vice President Xi" or even "President Xi".

A roar went up when his car rushed by and Xi waved his hand out the window.

"It was him for sure," said one student, who had taken a blurry shot of the car on his smart phone. "He must be better." The student refused to give his name.

The news spread rapidly on China's popular Twitter-like microblogging site Sina Weibo, with users referring to Xi as the "crown prince" to avoid the usual censorship associated with the names of top leaders.

"He looks well," wrote one user.

"In the future he should take better care when he goes swimming," added another.

Xi had been out of the public eye for almost two weeks and had skipped meetings with foreign leaders and dignitaries, including U.S. Secretary of State Hillary Clinton.

Chinese government officials repeatedly refused to say what had happened to him, fuelling speculation that has included Xi supposedly suffering a heart attack, a stroke, emergency cancer surgery and even an attempted assassination.

The health of the country's leaders has long been considered a state secret in China.

The ruling Communist Party's refusal to comment on his disappearance from public view and absence from scheduled events was in keeping with its traditional silence on the question of the health of top leaders, but it had worried or mystified most China watchers.

Xi had last appeared in public on September 1. He pulled a back muscle while swimming shortly before Clinton arrived on an official visit on September 4, the sources had said, forcing him to scrap a meeting with her the next day and also with Singapore Prime Minister Lee Hsien Loong.

Beijing has yet to announce formally a date for the party's five-yearly congress, at which Xi is tipped to replace Hu Jintao as party chief, although it is still expected to be held in mid or late October at the earliest.

In March next year, he is formally to take over the reins of the world's second-largest economy.

The uncertainty surrounding Xi's absence has had no impact so far on Chinese or foreign markets, which have been absorbed by Europe's debt crisis and China's own economic slowdown. But investors have been keeping a close eye on the mystery surrounding Xi, after months of political drama in China.

Senior leader Bo Xilai was suspended from the party's 25-member Politburo in April and his wife convicted of the murder of a British businessman. Blind human rights activist Chen Guangcheng escaped from house arrest in April and took refuge in the U.S. embassy before leaving for New York.

In another scandal this month, a senior ally of President Hu was demoted after sources said the ally's son was killed in a crash involving a luxury sports car.

LOS ANGELES (AP) ? A Southern California filmmaker linked to an anti-Islamic movie inflaming protests across the Middle East was interviewed by federal probation officers at a Los Angeles sheriff's station but was not arrested or detained, authorities said early Saturday.

Nakoula Basseley Nakoula, 55, was interviewed at the station in his hometown of Cerritos, Calif., Los Angeles County Sheriff's Deputy Don Walker said.

Federal officials are investigating whether Nakoula, who has been convicted of financial crimes, has violated the terms of his five-year probation. If so, a judge could send him back to prison.

Nakoula went voluntarily to the station early Saturday morning, wearing a coat, hat, scarf and glasses that concealed his appearance. It is unclear whether he returned to his home, which has been besieged by media for several days.

The probation department is reviewing the case of Nakoula, who pleaded no contest to bank fraud charges in 2010 and was banned from using computers or the Internet or using false identities as part of his sentence.

Federal authorities have identified Nakoula, a self-described Coptic Christian, as the key figure behind "Innocence of Muslims," a film denigrating Islam and the Prophet Muhammad that ignited mob violence against U.S. embassies across the Middle East.

Much of the film was shot inside the offices of Media for Christ, a nonprofit based in the Los Angeles-area city of Duarte. The charity raised more than $1 million last year "to glow Jesus' light" to the world.

The Riverside County man who was a script advisor to the film and who has a long history of anti-Islamic activism told the Press-Enterprise newspaper that he has received multiple death threats.

"I'm really tired," Steven Klein said when he answered the door of his home in Hemet, Calif., Friday with a pistol in his hand and clad only in a pair of white shorts stained with what appeared to be ink spots.

The newspaper said Klein, a Vietnam veteran, appeared agitated. While waving the gun, he told the newspaper he was standing up for his First Amendment rights in helping with the film and said he is prepared to die for those rights.

A federal law enforcement official said authorities had connected Nakoula to a man using the pseudonym of Sam Bacile who claimed earlier to be writer and director of the film.

Violent protests set off by the film in Libya played a role in mob attacks in Benghazi that killed U.S. Ambassador Chris Stevens and three other American officials. Demonstrations against American missions have since spread to several other countries.

Japanese, Chinese ships exchange island warnings

TOKYO (AP) ? Chinese and Japanese government ships exchanged warnings Friday in waters near disputed islands in the East China Sea, while Tokyo called on Beijing to protect its citizens amid anti-Japan protests and reported assaults in China.

Tensions between the Asian giants have flared anew after the Japanese government bought the islands from their private Japanese owners this week. The uninhabited islands, claimed by both countries as well as Taiwan, have become a rallying point for nationalists on both sides.

In response to Japan's purchase, China on Friday sent six surveillance ships into what Japan says are its territorial waters around the islands, called Senkaku by Japan and Diaoyu by China. Japanese coast guard ships radioed warnings to the Chinese vessels and two or three moved out of the territorial waters, said Yasuhiko Oku, a Japanese coast guard official.

Japan controls the islands, which are surrounded by rich fishing grounds and are near key shipping lanes, but China doesn't recognize those claims. State-controlled China Central Television repeatedly played footage of a Chinese Marine Surveillance officer aboard one of the ships radioing the Japanese vessels to demand they leave.

"The actions of your ships violate China's sovereignty and rights," the officer was shown saying. "Any unilateral act from your side regarding the Diaoyu islands and its affiliated islands is illegal and invalid. Please stop any infringing acts. Otherwise, your side will bear the consequences caused by your actions."

With a typhoon approaching the area, by Friday afternoon all six Chinese ships had left the 24-mile zone around the islands, said Yoshiyuki Terakado, another Japanese Coast Guard official.

Emotions have been running high since April, when Tokyo's nationalistic governor, Shintaro Ishihara, proposed buying and developing the islands so that they wouldn't fall into Chinese hands. Activists from both sides landed on the islands in August.

To block Ishihara's plan, which would have infuriated China, Prime Minister Yoshihiko Noda's government was left with little choice but to buy the islands. The government doesn't plan to develop them, but the move has still angered China, and Beijing has warned of "serious consequences."

Anti-Japanese protests have since been held in various Chinese cities, and state media has published calls for a boycott of Japanese goods.

The dispute has stirred up emotional memories of Japan's brutal occupation that ended only at the close of World War II. While Japan routinely apologizes for its wartime actions, its politicians often anger China by visiting the Yasukuni Shrine, a memorial to Japan's war dead, including top war criminals.

The Japanese Consulate in Shanghai reported on its website that several Japanese have been assaulted or harassed in the past few weeks. It said Chinese have thrown water bottles and hurled insults at Japanese walking on the street. One person was hit with soda by a Chinese person who shouted "Japanese!" A consular official said more than four people had been hurt in anti-Japanese attacks in the Shanghai area.

Japanese Chief Cabinet Secretary Osamu Fujimura called on Beijing to take steps to assure the safety of Japanese tourists and residents in China.

Chinese Foreign Ministry spokesman Hong Lei said while Chinese were upset with "violations of Chinese sovereignty," they had no problem with "Japanese people in general." He urged Chinese to "express demands legally and reasonably."

While visiting Australia, Japanese Foreign Minister Koichiro Gemba told reporters that "we should never let the situation escalate," and expressed "strong hopes for the Chinese government to respond to the situation in an appropriate and also a calm manner."

Officials both in Tokyo and Beijing maintained that they had the right to send ships to the islands.

China's foreign ministry said its decision to send its ships was part of legal "law enforcement and patrol activities aimed to demonstrate China's jurisdiction over the Diaoyu Islands."

Fujimura called the fleet's deployment an unprecedented violation of Japanese territory and "extremely regrettable." Japan summoned China's ambassador to lodge a protest.

Chinese Assistant Foreign Minister Le Yucheng said Japanese authorities have let the situation escalate by appeasing and giving free rein to 'trouble-making right-wing forces' at home.

Le catalogued incidents earlier this year that he said were provocations, including a fishing trip to waters around the islands by Japanese lawmakers and a visit to the islands by right-wing activists to mourn war dead.

"Japan's 'purchase of the islands' is by no means an isolated event," Le told more than a dozen Chinese scholars at a symposium on the history of the islands held in a Beijing government compound. "It is a result of the changing political climate in Japan. There is a sinister tendency inside Japan that is taking Japan and China-Japan relations down an extremely dangerous road."

TUNIS, Tunisia (AP) ? Violent protests outside the U.S. Embassy in Tunis against an anti-Muslim film were met with tear gas and gunshots Friday, leaving two people dead, around 40 others injured and plumes of black smoke wafting over the city.

Several dozen protesters briefly stormed the U.S. Embassy compound in Tunisia's capital, tearing down the American flag and raising a flag with the Muslim profession of faith on it as part of the protests. Protesters also set fire to and looted an American school adjacent to the embassy compound and prevented firefighters from approaching it. The school appeared to be empty and no injuries were reported.

Earlier, several thousand demonstrators had gathered outside the U.S. Embassy, including stone-throwing protesters who clashed with police, according to an Associated Press reporter on the scene. Police responded with gunshots and tear gas. Police and protesters held running battles in the streets of Tunis. Amid the unrest, youths set fire to cars in the embassy parking lot and pillaged businesses nearby.

The state news agency TAP, citing the health ministry, said both of those killed were demonstrators, while the injured included protesters and police. Two of the injured were in critical condition, the health ministry said.

A Tunisian employee of the U.S. Embassy who had an injured leg was taken out on a stretcher to an ambulance. It wasn't immediately clear if there were any other injuries. Embassy officials did not respond to calls and emails.

The group that breached the U.S. Embassy's outer wall was eventually pushed back outside by a huge deployment of police and special forces. As night fell, the crowd of protesters outside the embassy dwindled to a handful.

The al-Wataniya 1 television station said the presidential guard also intervened and escorted the U.S. ambassador and about 80 embassy personnel away from the site to safety.

Crowds angry over an anti-Muslim film ridiculing the Prophet Muhammad have assaulted U.S. embassies across the Middle East.

The degree of violence in Tunisia surprised many and raised new questions about the direction of the country, where an uprising last year forced out its longtime president and set off pro-democracy revolts across the Arab world. A once-banned Islamist party came to power in elections last year, but the moderate government has struggled to quell protests by increasingly vocal ultraconservative Muslims known as Salafis.

US won't rule out militant link to attack in Libya

The deadly attack on the U.S. Consulate in Libya was staged by militants who set the building on fire. NBC's Richard Engel reports.

Updated at 7:08 p.m. ET: U.S. officials told NBC News on Wednesday that they couldn't rule out the possibility that al-Qaeda-inspired Islamist militants were responsible for the attack in the eastern Libyan city of Benghazi?that killed Christopher Stevens,?the U.S. ambassador to Libya, and three other Americans.

U.S. and Libyan officials, independent analysts and postings on Islamist websites from known militant activists suggested that the attack ? which officials had previously suggested was retaliation for release of a movie critical of the Muslim Prophet Muhammad ? may have been an orchestrated assault, noting that it occurred on the 11th anniversary of the Sept. 11, 2001, terrorist attacks in the U.S.

The State Department ordered?all "non-emergency" U.S. personnel out of the country, but?officials told NBC News that as of now there was no plan for a total U.S. evacuation.

U.S. officials said the Navy is positioning two guided-missile destroyers off the Libyan coast to provide "additional military options" if needed.

The USS LaBoon had already arrived off the coast of Libya, officials told NBC News. The USS McFaul was expected to arrive Thursday. Both are based at Norfolk, Va., and are listed as having crews of 338.

In addition, the Defense and State departments were considering sending additional Marines to other potential trouble spots, including Cairo and Kabul, which have both seen sectarian flare-ups against the U.S.

At a briefing for reporters, Libya's interim president, Mohammed el-Megarif, suggested that the attack was timed to coincide with the anniversary specifically to "destroy democracy in Libya."

Al-Qaeda leader Ayman al-Zawahiri released a video Tuesday marking the anniversary. In it, he called for Islamists to seek revenge for the killing by a U.S. drone strike of deputy al-Qaeda leader Abu Yahya al-Libi in June.

Online postings Wednesday from Islamist militants were celebrating the attack as payback for his death, said Evan Kohlmann, an Middle East and terrorism analyst for NBC News.?One of the postings claimed responsibility for the attack in the name of Ansar al-Sharia, ???a Sunni Muslim militant faction.

Noman Benotman, a Libyan-born senior strategy analyst for the Quilliam Foundation, an antiterrorism research group in London, also said the organization's intelligence indicated that the attacks were "committed by uncontrollable jihadist groups," not by a protest mob that spontaneously erupted into violence.

Quilliam said on its website that its sources, some of whom it said were in Benghazi itself, reported that about 20 militants carried out the attack using rocket-propelled grenade launchers.

That appeared to square with an eyewitness account cited by Libya Herald, which quoted a demonstrator at the consulate as saying a protester fired a rocket-propelled grenade launcher at a police vehicle but hit the consulate building on Venezia Street, instead.

The Voice of America, meanwhile, cited similar reports that "several dozen gunmen from the Islamist group Ansar al Sharia" may have carried out the attack.

Tommy Vietor, a spokesman for the National Security Council, said "it would be premature to ascribe any motive to this reprehensible act," but other U.S. officials said they couldn't rule out the possibility of a link to al-Liby's death or the Sept. 11 anniversary, saying the attack was too sophisticated to have been spontaneous.

In the aftermath, officials told NBC News that they were preparing to send as many as 200 Marines to Libya to bolster security around the U.S. Embassy in Tripoli. The Marines' Fleet Anti-Terrorism Security Team would be deployed from a Marine Amphibious Ready Group already positioned aboard a helicopter carrier in the North Arabian Sea, the officials said.

Obama condemns attack
President Barack Obama condemned the attack, in which the Americans were killed after protesters stormed the U.S. consulate Tuesday night in Benghazi.

President Barack Obama vowed to bring the killers of the four Americans to justice. NBC's Andrea Mitchell reports.

"Make no mistake: Justice will be done," Obama said Wednesday.

Stevens, 52, was the first U.S. ambassador to be killed during an assignment since Adolph Dubs was slain in an exchange of gunfire during a kidnapping attempt in Afghanistan in 1979.

The State Department identified a second victim as Sean Smith, a Foreign Service information management officer and father of two children. Smith had previously been posted to the U.S. Consulate in Montreal, the U.S. Mission in Canada confirmed.

The State Department said the families of the two other victims were still being notified. U.S. officials would say only that they were employed by the State Department.

Ambassador Stevens was 'courageous and exemplary,' Obama says

"While the United States rejects efforts to denigrate the religious beliefs of others, we must all unequivocally oppose the kind of senseless violence that took the lives of these public servants," Obama said in a statement.

Vice President Joe Biden, meanwhile, insisted the U.S. wouldn't be driven from the country.

"We never have been, and we never will be, run off, period," Biden said at a campaign event in Dayton, Ohio. "That's not who we are."

Secretary of State Hillary Clinton said Wednesday that the attack "should shock the people of all faiths around the world."

"I ask myself, how could this happen? How could this happen in a country we helped liberate, in a city we helped save from destruction?" she said. "This question reflects just how complicated and, at times, how confounding the world can be."

U.S. Muslims said they were concerned about possible retaliatory attacks on their mosques, the Washington-based Council on Arab-Islamic Relations said Wednesday as it also condemned the attacks.

"We're starting to get hate calls, and we'd already seen a wave of anti-Muslim incidents," Ibrahim Hooper, a spokesman for CAIR in Washington, told NBC News. "Muslims have to feel insecure."

Descriptions of Tuesday's attack depicted chaos and bloodshed, with Libyan security overrun and retreating.

Mohammed el-Megarif, Libya's interim president, apologized to the U.S. for the deadly attack. ITV's Richard Pallot reports.

Stevens died of severe asphyxiation, apparently from smoke, Ziad Abu Zeid, a Libyan doctor who treated him, told The Associated Press.

Stevens was practically dead when he arrived close to 1 a.m. Wednesday (7 p.m. ET Tuesday), and "we tried to revive him for an hour and a half but with no success," Abu Zeid said. The ambassador was bleeding in his stomach because of the asphyxiation but had no other injuries, he said.

Demonstrations in Benghazi and in Egypt broke out Tuesday following news reports about the anti-Islamic video, which ? if a trailer posted on YouTube is representative ? features an amateur cast performing a wooden dialogue of insults disguised as revelations about Muhammad.

The diplomatic crisis in Libya and Egypt quickly turned political as the Obama and Romney campaigns traded statements overnight. NBC's Chuck Todd reports.

The attacks were the first such assaults on U.S. diplomatic facilities in either country, at a time when both Libya and Egypt are struggling to overcome the turmoil following the ouster of their longtime leaders, Moammar Gadhafi and Hosni Mubarak, in uprisings last year.

The events appeared to underscore how much the ground in the Middle East has shifted for Washington, which for decades had close ties with Arab dictators who could be counted on to muzzle dissent.

Thunderstorms delay flights, snarl traffic in Vegas area, flood homes in Southern California

LAS VEGAS, Nev. - Intense thunderstorms drenched parts of the Southwest on Tuesday, delaying flights and stranding motorists in the Las Vegas area and flooding two mobile home parks in Southern California.

East of downtown Las Vegas, television news video showed yellow school buses inching slowly along swamped roads in some neighbours and muddy brown water up to the lower window sills of stucco homes in others.

A Twitter photo showed dozens of cars submerged in water up to their headlights in a parking lot outside a University of Nevada, Las Vegas sports arena.

The National Weather Service issued severe thunderstorm and flash-flood warnings before and after almost an inch of rain was reported at McCarran International Airport just before 2 p.m.

Departures were postponed and arrivals were delayed after the airport ordered a stop on fueling operations during lightning strikes, airport spokeswoman Linda Healey said.

Firefighters responded to more than 20 calls about people in stalled cars, county spokesman Dan Kulin said. A Las Vegas police helicopter was dispatched during the height of the storm to pluck several people from swamped vehicles on area roadways, Officer Bill Cassell said.

After responding to numerous 911 calls, officials in Clark County, North Las Vegas, Henderson and Las Vegas said Tuesday there were no confirmed reports of serious injuries.

National Weather Service meteorologist Michael Staudenmaier said more than 1.75 inches of rain were reported in downtown Las Vegas.

The rainfall amounts put the region on pace to exceed the 4.5 inches of rain it normally gets in a year, he said.

However, National Weather Service meteorologist Scott Sukup said the Nevada showers weren't part of the same storm system that doused parts of Southern California.

There, a thunderstorm that dropped more than the average annual rainfall on parts of the Coachella Valley in one night alone caused flooding at two mobile home parks, forced road closures and dampened a school, officials said Tuesday.

The early morning thunderstorm stalled for six to eight hours over Mecca and Thermal, two towns at the southern tip of the Coachella Valley 150 miles southeast of Los Angeles. Thermal is about eight miles from Indio, Calif., where the annual Coachella Music Festival is held.

The storm dropped 5.51 inches of rain near Mecca and 3.23 inches of rain near Thermal, meteorologist Mark Moede said. The average annual rainfall in Thermal is just shy of 3 inches, he said.

"That's an amazing amount of rain," Moede said. "It's unusual anywhere to get a storm that sits stationary for five to eight hours. The fact that it occurred in the southern part of the Coachella Valley is even more unusual because it's typically a very arid part of the country."

In Thermal, the downpour flooded the Desert Mobile Home Park better known as Duroville, causing the park to lose one of its wells and creating concerns about overflow from sewage ponds used to treat waste, said Ray Smith, a Riverside County spokesman.

County fire crews assessed the water and electrical systems and were providing water to the park. Residents also were advised to boil water, he said.

Several Duroville homes remained without power late Tuesday afternoon. About 1,400 people live in the park, but it wasn't immediately clear if any evacuated due to the flooding. A voluntary evacuation centre was established in Mecca.

St. Anthony's Mobile Home Park in Mecca also was affected, Smith said, but fared better than Duroville. Video clips showed Mecca residents wading through streets with water reaching their knees and cars creeping through flooded residential streets.

Flooding also was reported at Mecca's Saul Martinez Elementary School, but students doubled up in some classes and the school remained open, The Desert Sun newspaper reported.

Meanwhile, some southern Utah residents also were dealing with flooding because of a broken dike.

Officials in Santa Clara were inspecting whether people could return to more than 30 homes and businesses that were evacuated after the Tuesday break flooded nearly four square blocks.

City Manager Edward Dickie said the dike along a retention pond broke open after heavy morning rains, sending a deluge of water into downtown.

"It didn't just breach. It broke. It's gone," he said, adding that the flooding quickly receded as water drained into rivers and creeks.

Some streets through town remained closed late in the day as authorities assessed the damage and waited for floodwaters to recede. No injuries were reported.

Crews used sandbags to shore up the broken dike and stave off the potential for additional flooding if the rain returns.

Researchers have created a new soft appendage made from flexible plastic, that uses three compartmentalized air channels to achieve a snake-like range of movement. The touch of the tentacle is so light, that it is able to pick flowers without damage. While suggested applications include working with fragile objects, or in hard to reach locations, the team also experimented by adding cameras, suction cups and -- most terrifyingly -- syringes to the tip. The only limitation, apparently, is that the air channels prevent it from being scaled down. So while our insect friends are safe from strangle-bot, we might not be so lucky.

Failed Alzheimer's drug showed signs of working: studies

CHICAGO (Reuters) - New data on Pfizer Inc and Johnson & Johnson's Alzheimer's drug, bapineuzumab, show the treatment reduced underlying markers of the disease in some patients, suggesting the failed medication might work at an earlier stage.

The findings from two large studies, presented at a European neurology meeting in Stockholm on Tuesday, followed the companies' announcement last month that they were scrapping large-scale clinical trials of the drug after it failed to improve memory or thinking skills in patients with mild to moderate Alzheimer's.

Many researchers had expected bapineuzumab to fail this test because they believe that Alzheimer's starts years before memory problems become apparent, and that treatment when patients already have dementia may be too late.

But they have been eagerly awaiting the so-called biomarker results, which measure fluids and tissues in the body, to see if the drug hit its biological targets and therefore, could work at an earlier stage of the disease.

The biomarker results show that bapineuzumab significantly reduced levels of the protein beta amyloid on the brain scans of patients with a gene mutation that increases their risk of Alzheimer's, compared with subjects who were give a placebo.

The drug also significantly reduced the amount of a toxic form of the protein tau in spinal fluid, a sign of brain cell death, compared with patients who were given a placebo.

However, MRI tests showed patients in the treatment and placebo groups had a similar loss of brain volume.

Dr. Reisa Sperling of Harvard Medical School and Brigham and Women's Hospital presented results of a study of patients who carry the ApoE4 gene mutation. She said the findings were not a "home run in biomarkers," but are encouraging.

"I am happy there is evidence that we are having some effect on the disease process in the brain," she said in a telephone interview from Stockholm.

"I am very interested in moving towards a much earlier stage of Alzheimer's disease in which we might be able to impact the biology at a time when we might prevent symptoms."

The results of a second trial of patients who were not carriers of the ApoE4 mutation, presented by Dr. Stephen Salloway of Brown University in Rhode Island, showed no significant improvements, but there was some reduction in a marker of brain cell death at the highest dose. Because the follow-up study only included 39 patients, however, it was likely too small to show much of an effect, Salloway said.

In both trials, a significant number of patients experienced a brain-swelling side effect known as vasogenic edema, and rates of this problem increased as the dose of the drug increased.

In 2009, the highest dose of the drug was dropped from the non-carrier study because of this brain-swelling issue.

Researchers said the lower doses tested in the studies -- 0.5 milligram per kilogram in the ApoE4 carriers and both 0.5 mg/kg and 1.0 mg/kg in the non-carriers -- may explain why the patients failed to see a benefit in memory and thinking skills.

'POSITIVE DIRECTION'

Maria Carrillo of the Alzheimer's Association, who had seen the results, said they suggest the biomarkers were moving in a "positive direction."

Sanford C. Bernstein analyst Tim Anderson said in a note to clients the results "leave open the possibility" that the bapineuzumab may work in earlier stage disease.

Researchers will be comparing these findings to those of Eli Lilly and Co's similar drug, solanezumab. Although the Lilly drug also failed to meet its study's main goals in mild to moderate patients, the company did see a slight benefit when they isolated results for mild patients.

Biomarker results for Lilly's drug are expected to be released on October 8 at a neurology meeting in Boston. Researchers have not yet analyzed pooled data on mild patients in the bapineuzumab studies.

Although Pfizer and J&J discontinued all studies involving the intravenous form of bapineuzumab, they have an ongoing study testing the treatment in a shot form.

Dr. Eric Yuen of J&J said the newer formulation may offer a more smooth exposure to the drug than the IV version, which could help reduce some of the side effects.

Sperling said side effects will play a role in the future of bapineuzumab. "I think it will be important to see whether we will get a better side effect profile that will allow us to dose higher," Sperling said.

Several studies, including one proposed by Sperling, will be testing drugs such as bapineuzumab in patients with normal memories who are likely to develop Alzheimer's, either because they are genetically predisposed to it or because biomarker tests suggest it is already forming.

Salloway, who is also helping to organize a study known as DIAN in patients with an inherited form of the disease, said researchers will examine all available data in order to pick compounds likely to influence the disease process early on.
